Starbucks Corporate Strategy Essay Starbucks Corporate Strategy Corporate Strategy fundamentally is concerned with the selection of businesses in which the company should compete and with the development and coordination of that portfolio of business. [1] In the case of Starbucks the corporate strategy they have implemented is unique to their industry which has allowed them to differentiate from their competitors and is summarized best by Howard Schultz CEO of Starbucks, â€Å"We’re in the people business serving coffee,[2]† high quality specialty coffee and related products in a European cafe environment. It is clear Starbucks is in a growth strategy utilizing three key techniques that support its Mission, â€Å"to inspire and nurture the human spirit – one person, one cup and one neighbourhood at a time†[3]. Vertical Integration Tying in with Starbucks’ Mission, the companies vertical integration with its supply chain from roasting its own coffee in one of two of its plants to its involvement with developing and fostering strategic partnerships with local coffee farmers may be the most important growth strategy the company focuses on. Quality and corporate social responsibility are the pillars of Starbucks; vertical integration is the key component to insuring both of these fundamental characteristics of Starbucks are managed. Diversification Starbucks has created related diversification into wholesale distribution by developing coffee based branded products with Pepsi-Cola and Dreyer’s Inc. that can be sold in grocery stores. The company is also expanding into non-related industries by creating an entertainment division focusing on the music and movie industry. As Starbucks expands diversification will become as important as its vertical integration as it moves through the product lifecycle in the coffee industry. (See Appendix A) Horizontal Integration Starbucks has expanded with horizontal integration by acquiring Seattle’s Best Coffee and Torreazione Italia Coffee. With the increased competition and new entrants in the coffee industry horizontal integration may become a greater focus for Starbucks. Starbucks Business Strategy At the business level, the strategic issues are less about the coordination of operating units and more about developing and sustaining a competitive advantage[4]. In order to understand the Coffee Industry’s competitive advantage the use of Porters five Competitive Forces Model should be utilized. (appendix b) Starbucks has gained its competitive advantage by the use of diversification. The company has focused, through vertical integration corporate strategy, on providing the highest quality coffee products in a sustainable practice that is highly identifiable by its customer base. The use of their innovative marketing design with their European Cafe feel and creative diversification with its products has allowed it to be classified as, â€Å"the most dynamic retail brand conceived over the last two decades†[5] Starbucks faces a high degree of rivalry and a medium degree of buyer power (appendix b). The coffee industry also retains many characteristics of having high Consumer Market Segmentation, which can be supported by the relatively large span in the price for a cup of coffee and the varying degree in which certain coffee chains are found in certain particular geographic regions and outlets. Starbucks has found a way to incorporate a broad strategy attracting a broad consumer despite the high prices of its products[6]. This supports that their differentiation strategy is affective and it is clear consumers can identify the high quality of their products and the overall experience of visiting one of their locations. With a high threat of new entrants (appendix b) and the presence of high Consumer Market Segmentation, Starbucks’ strong differentiation strategy will allow it to remain competitive in light of this high threat.

Purify GFP From Aequorea Victoria

Purify GFP From Aequorea Victoria Methods and Results: GFP was cloned into E. coli strain JM109 and expressed under optimal conditions in Luria broth agar containing ampicillin and IPTG for induction. Protein was extracted by lysis using bead milling technique and fluorescence of protein measured in a fluorimeter, concentration of both pure and crude proteins were obtained with Bradford (1976) method. Purity of GFP was further confirmed by SDS-PAGE stained with coomassie blue. Conclusion: Specific activity(RFUmg-1) of pure protein increased compared to crude representing increase in purity, with a substantial yield of 82%. Significance and Impact of Study: This study proved Ion exchange chromatography as a reliable technique for GFP purification and high percentage recovery for use as a reporter gene in molecular biology studies. INTRODUCTION The jelly fish Aequorea victoria, emits a bluish light from the margin of its umbrella (Inuoye, and Tsuji 1994). The light is produced by the bioluminescent jellyfish when calcium binds to the photoprotein aequorin. Although activation of aequorin in vitro or in heterologous cells produces blue light, the jelly fish produces green light. This light is the result of a second protein in A. victoria that derives its excitation energy from aequorin, the green fluorescent protein (Chalfie et al., 1994). Green fluorescent protein (GFP) is a protein of 238 amino acid residues. It is a highly stable protein possessing a tightly packed ÃŽ ²ÃƒÅ'†°ÃƒÅ'†°ÃƒÅ'†°ÃƒÅ'†°can tertiary structure that is resistant to many biological denaturants, most proteases, pH (5-12), temperature (Tm=78  °C), and chaotropic salts (McRae et al., 2005; Zhuang, et al., 2008). Purified GFP absorbs blue light (maximally at 395nm with a minor peak at 470nm) and emits green light (peak emission at 509nm with a shoulder of 540nm). This fluorescence is stable and virtually no photobleaching is observed (Chalfie et al., 1994). The stable and intense fluorescence of GFP without any cofactors in many different organisms makes is ideal for molecular biology applications such as markers for gene expression analysis of molecular interactions and also as biological information storage devices and optical biosensors in areas of Nanotechnology (McRae et al., 2005). Current purification procedures, specific for GFP include multiple phase high performance liquid chromatography (HPLC), chromatofocusing on a pH gradient, metal ion precipitation and organic extraction. Teen Smoking Teen smoking. Those two words mean a lot of things to a lot of different people. To some it means nothing. They are just two meaningless words found under T and S in the dictionary. To others it is as if these words symbolize some sort of treason or crime against society. Yet to others it is just another stereotype to be placed under. And to some it is a salvation. An escape. Unfortunately I am writing this paper so you will get my positions on teen smoking rather than other peoples. Teen smoking gathers a lot of emotions when I think about it. It makes me very angry sometimes. My first draft of this was good but I saw myself getting angry and lashing out on everyone, so I revised it. But enough of that, I find teen smoking to be a very controversial subject. To be completely honest with you I am smoking right now as I am writing this. Obviously I am a smoker. I am 17 years old and I am a smoker. In the late 1990’s, the statistics showed that approximately 25% of teens smoke. That’s one out of every four teenagers. High school is a tough time for teens. These years are critical to a teens future. This explains why a vast majority of smokers start at 16 years or younger with the most common age being 14 years old(freshman). It has also been proven also that teens who score lower in school smoke more than higher scoring students do. It seems that everyone smokes in our school. Our school is overrun with smokers. It is right now at least 50% smokers and 65% if you count the people who will smoke before their high school career is up. We practically encourage it. I mean Fireman’s Field practically condones teen smoking. Teen smoking is defiantly a problem in our school, as well as schools all over the United States. I feel that teen smoking is a huge problem. I feel that too many teenagers smoke cigarettes. It is something that needs to get dealt with. But before I get into that I should probably start with the causes of teen smoki ng. There are several factors that start teens on smoking cigarettes. There has to be considering that over 1 million new teenagers will start smoking annually. The most common is peer pressure. Depth Perception, an Inborn Skill? :: essays research papers

In 1960, Eleanor Gibson and Richard Walk conducted an experiment to see whether depth perception is an inborn or a learned skill in humans. They conducted their experiment with a table that had a thick glass surface on half of the table and a solid base on the other half. This created an illusion of a small cliff without the dangers of actually falling. In this experiment, infants ranging from the age of 6 to 14 months were placed on the solid side of the table. The infants’ mothers were placed on the other side of table and were there to coax the infants to the other side. Of the 30 infants tested, 27 of them crossed the glass surface when called while only 3 refused.   Ã‚  Ã‚  Ã‚  Ã‚  Gibson and Walk conducted the same experiment on newborn chickens and goats with astonishing results. When chickens and goats were placed on the solid side, not a single one of them made an error to cross the â€Å"cliff.† The same test was conducted on baby rats whose results fared far worse than the results of the chickens and goats. The rats fared worse because they are nocturnal animals who rely on other senses other than vision to direct them. From this experiment, Gibson and Walk concluded that depth perception was inborn to all animals and humans by the time they achieve independent movement. This is in the case of chickens and goats at birth and for humans at around 6 months of age.   Ã‚  Ã‚  Ã‚  Ã‚  The results of Gibson and Walk’s experiment are very questionable because their control group did not consist of any socially dependant animals. Infants are socially dependent of their mother for survival and nurturing throughout their childhood. Gibson and Walk should have conducted their control experiment on socially dependant animals such as elephants or cheetahs instead of animals that do not rely heavily on their caretaker. In 1985, Sorce, Emde, Campos, and Klinnert conducted the same visual cliff experiment with human infants and their mothers. This time, the mother was instructed to maintain an expression of fear or happiness on the other side of the â€Å"cliff.

Dr. Henry Jekyll was born in to a society of morality, respectability and religion. It was believed that ‘progress’ could only be made if everyone was self-disciplined and moralistic. Authors such as Samuel Smiles wrote â€Å"Self-Help† guides. All this was aimed to help the rich get richer and the poor get poorer. In an age of stern, industrious hypocrites, respect was everything to the upper and middle classes. People denied themselves alcohol, gambling and prostitution to gain the respect of others. Jekyll refers to these denied as â€Å"pleasures†. People lived without these â€Å"pleasures† but soon began to wonder what they were missing. This brought about the â€Å"slum adventurers†. These were middle and upper class men who wanted to keep the respect of their society but, through anonymity, still indulge in the â€Å"pleasures† the poor slums had to offer. They would work by day in their offices and at night would journey down t o the alleys of the slums. A person such as Mr Utterson, a London lawyer who does not wish to indulge in pleasures is of a vicarious nature. It is noticed that â€Å"†¦though he enjoyed the theatre, [he] had not crossed the doors of one in twenty years†. It is this vicariousness that helps him solve the case of Dr. Jekyll and Mr Hyde. Utterson is the opposite of Jekyll in the way that Utterson keeps the respectability of the Victorian society, whereas Jekyll ‘rebels’. The rebellious nature of Jekyll leads him to discover how to transform his appearance. Dr. Lanyon, a respectable conventional doctor of Victorian society frowns on Jekyll mostly secret work, which he refers to as â€Å"unscientific balderdash†. In the beginning it is difficult for us to feel sympathy for Dr. Jekyll: he is acting by his own conscience. At this current stage, he is in no way addicted to Mr. Hyde. Even the sight of Mr. Hyde â€Å"†¦ pale and dwarfish†¦ who gave the impression of deformity but with no nameable malformation†, according to Mr. Utterson, who â€Å"†¦had taken a loathing to my gentleman at first sight†, should have forced Jekyll to recognise that Hyde was evil. It is very difficult to feel sympathy for Jekyll after we are told about when Hyde â€Å"†¦trampled calmly over†¦Ã¢â‚¬  a young girl’s body. Normally after this event anyone else would have ceased his transformation into Hyde. But Jekyll starts to become addicted to taking the drugs (as with modern addictions) and continues to turn into Hyde even after trampling the girl. As if trampling the girl was not a big enough deterrent Hyde brutally murders Sir Danvers Carew. Jekyll, knowing what had happened, easily accepts it and shifts the blame to Hyde. He shows this in Dr Jekyll’s full statement of the case, â€Å"it was Hyde, after all, and Hyde alone that is guilty†. The ease at which he accepts this and shifts the blame could almost be inhuman. But after he discovers that he begins to change into Hyde spontaneously, without the drug we see he is very self centred and selfish, â€Å"I must have stared upon it half a minute†¦I rushed to the mirror†¦ my blood was something exquisitely thin and icy†. Even though Jekyll is selfish about this, not mentioning what a terrible thing that he made now controls. We can feel some sympathy for knowing that he is trapped and at anytime Hyde might emerge over which Jekyll has no control. Alternatively we can have sympathy for a man that failed to have foreseen what would happen and stopped the process as soon as possible. But he continued to satisfy his need for the â€Å"pleasures† which otherwise eluded him. It is also very difficult to feel sorry for someone who knows and remembers what happens, but does not take action to prevent it. â€Å"My two natures had memory in common†. Surely the memories of the girl and Carew should be painful enough to force any sane and humane person to put a stop to Hyde’s actions. Hyde goes into hiding after Carew’s murder in fear of being hung, should he be caught. This removes a lot of the sympathy we may otherwise feel towards Jekyll at this point, as yet again he shows signs of being selfish and putting himself before everyone else. Some of this sympathy is regained when he is in hiding, he begins to show genuine remorse for Carew’s death. In the statement of the case, Jekyll finally admits to Hyde’s evil by saying â€Å"†¦It was no longer the fear of the gallows, it was the horror of being Hyde that racked me.† This shows that Jekyll has become altruistic. Jekyll has now begun to put himself after everyone. He now excepts the he must not let Hyde free again for fear of him committing more evil. It is now we finally begin to feel sympathy and start to respect Jekyll for trying to put a stop to Hyde. Finally Jekyll commits suicide to save the world from the terror and evil Hyde could unleash. This gains him the greatest respect and sympathy. He took his life to save others from the extreme evil he had inadvertently created. He finally takes ultimate responsibility and puts a stop to Hyde. In conclusion I think that Jekyll was a victim of Victorian society where respect was everything. In some parts it is hard to offer our sympathy but his final act was one that one must respect and offer sympathy for the pain Jekyll went through. As we have seen restricting things from people can only bring out the bad side of them, as Jekyll explains, â€Å"My devil had been long caged, he came out roaring†. Personal freedom is one of our greatest assets and one we take very much for granted.

Soil Behaviour and Geotechnical Modelling

(a) Discuss advantages and limitations of Duncan and Chang's model. Duncan and Chang's model assumes a hyperbolic stress-strain relation and was developed based on triaxial soil tests. The original model assumes a constant Poisson's ratio while the revised model accommodates the variation of Poisson's ratio by means of stress-dependent Poisson's ratio or stress-dependent bulk modulus. The Duncan-Chang model is advantageous in analyzing many practical problems and is simple to set up with standard triaxial compression tests. When tri-axial test results are not available, model parameters are also abundantly available in literatures. It is a simple yet obvious enhancement to the Mohr-Coulomb model. In this respect, this model is preferred over the Mohr-Coulomb model. However, it has its limitations, including, (i) the intermediate principal stress s2 is not accounted for; (ii) results may be unreliable when extensive failure occurs; (iii) it does not consider the volume change due to changes in shear stress (shear dilatancy); (iv) input parameters are not fundamental soil properties, but only empirical values for limited range of conditions. (v) the model is mainly intended for quasi-static analysis. (b) Discuss advantages and limitations of Yin and Graham's KGJ model. Yin and Graham's KGJ model is formed using data from isotropic consolidation tests and consolidated undrained triaxial tests with pore-water pressure measurement. It provides functional expressions for , , , and relationships in soils. In Duncan and Chang's model for triaxial stress conditions: may cause volume strain ( dilation and compression) may cause shear strain. Whereas Yin and Graham's KGJ model: Thus the volume change and shear strain was taken into account, which is an improvement to Duncan and Chang's model. The limitation of Yin and Graham's KGJ model may exist in the determination of the parameter and the complexity of its calculation. (c) Discuss the differences between elastic models and hypo-elastic models. For soils, the behaviour depend on the stress path followed. The total deformation of such materials can be decomposed into a recoverable part and an irrecoverable part. Hypoelasticity constitutes a generalized incremental law in which the behaviour can be simulated from increment to increment rather than for the entire load or stress at a time. In hypoelasticity, the increment of stress is expressed as a function of stress and increment of strain. The Hypoelastic concept can provide simulation of constitutive behaviour in a smooth manner and hence can be used for hardening or softening soils. Hypoelastic models can be considered as modification of linear elastic models. However, it may incrementally reversible, with no coupling between volumetric and deviatoric responses and is path-independent. 5.2 Use sketches to explain the physical (geometric) meaning of all 7 parameters (only 5 independent) in a cross-anisotropic elastic soil model (). In the multi-axial situation it is not sensible to talk about a yield stress. Instead, a yield function is defined which is a scalar function of stress and state parameters. (ii) A yield criterion, often expressed as yield surface, or yield locus, is an hypothesis concerning the limit of elasticity under any combination of stresses. There are two interpretations of yield criterion: one is purely mathematical in taking a statistical approach while other models attempt to provide a justification based on established physical principles. Since stress and strain are tensor qualities they can be described on the basis of three principal directions, in the case of stress these are denoted by , and . (iii) Potential surface is the segment of a plastic potential surface plotted in principal stress space, as shown in Figure 6.1 (a). A two dimensional case was shown in Figure 6.1 (b). When the material is plastic the stress state must satisfy the yield function. (b) Explain and discuss the associate flow rule and non-associate flow rule and how the two rules affect the volumetric deformation and the bearing capacity of a strip footing on sand. Sometimes simplification can be applied by assuming the plastic potential function to be the same as the yield function (i.e. ). In this case the flow rule is said to be associated. The incremental plastic strain vector is then normal to the yield surface and the normality condition is said to apply. In the general case in which the yield and plastic potential functions differ (i.e. ), the flow rule is said to be non-associated. If the flow rule is associated, the constitutive matrix is symmetric and so is the global stiffness matrix. On the other hand, if the flow rule is non-associated both the constitutive matrix and the global stiffness matrix become non-symmetric. The inversion of non-symmetric matrices is much more costly, both of storage and computer time. As noted, it occurs in a special class of plasticity in which the flow rule is said to be associated. Substitution of a symmetric for all elements in a finite element mesa, into the assembly process, results in a symmetric global stiffness matrix. For the general case in which the flow rule is non-associated and the yield and plastic potential functions differ, the constitutive matrix is non-symmetric. When assembled into the finite element equations this results in a non- symmetric global stiffness matrix. The inversion of such a matrix is more complex and requires more computing resources, both memory and time, than a symmetric matrix. Some commercial programs are unable to deal with non-symmetric global stiffness matrices and, consequently, restrict the typo of plastic models that can be accommodated to those which have an associated flow rule. (c) Explain plastic strain hardening and plastic work hardening or softening. The state parameters, , are related to the accumulated plastic strains .
